Molecular profiling of soft tissue sarcomas in tumor and peripheral blood in terms of response to treatment and early detection of recurrence or dissemination

Description

The project aims to utilize a basic molecular approach for monitoring soft tissue sarcoma (STS) by analyzing specific molecular alterations in circulating nucleic acids (ctDNA), along with personalized tumor tissue whole genome sequencing results, to address the critical need for biomarkers to predict and monitor patient response to therapy during preoperative treatment, identifying early recurrence or metastases during follow-up, and improving understanding of sarcoma-genesis for therapeutic advancements. The methodology involves profiling the genome of STS using droplet digital polymerase chain reaction (ddPCR) for detecting actionable mutations, monitoring cancer progression, and predicting therapy response.

The project also incorporates genetic changes analysis and gene expression profiling using next generation sequencing (NGS) and Oxford Nanopore Technology (ONT) in the primary tumor biopsy. Additionally, the

monitoring of circulating tumor-derived ctDNA (circulating tumor DNA, liquid biopsy) during treatment and posttreatment follow-up is proposed as a novel approach for assessing treatment response and detecting recurrence earlier than conventional imaging tests.
